Luke Macfarlane was born on 19th January 1980, in London, Canada as Thomas Luke Macfarlane Jr. His father Thomas Macfarlane was a director of student health services at the University of Western Ontario. While his mother, Penny Macfarlane worked as a mental health nurse.

Macfarlane grew up with a twin sister, Ruth Macfarlane, and an older sister Rebecca Macfarlane. When he was a child, he studied at the Lester B. Pearson School for the Arts and graduated from the London Central Secondary School. After moving to New York, he graduated from the Juilliard School in acting. 

In June 2018, he became an American citizen.
